A novel intumescent (carbonization, acid donor and foaming) fire retardant that mimics carbon nanotubes was introduced into bitumen roofing and characterized using cone calorimetry as the main analytical tool. The experimental results indicate that 18% (by mass) attapulgite mineral (ATTP) mixed with base bitumen decreased the peak heat release rate per unit area (pHRRPUA) by 10%. Further, incorporation of melamine coated ammonium polyphosphate (MAPP) decreased the pHRRPUA by 52% and a mixture of these (3:1, ATTP:MAPP) decreased the pHRRPUA by 25% as compared to adding CaCO3 as a filler. The residual mass loss after the cone test was also improved with up to 3%. The indication of a positive synergistic flame retardant effect of the ATTP-MAPP mixture is supported by thermogravimetric analysis. The addition of this rod-like mineral improved the general fire retardant properties of the base bitumen and increased the viscosity. Therefore, the polymer-modified bitumen with both fire retardant and rheological properties (providing mechanical strength) is a promising novel approach in the design of bitumen roofing membranes.  相似文献

对赤红球菌的组氨酸激酶基因进行密码子优化,将优化后的组氨酸激酶基因(rhks)构建重组表达质粒pGEX-4T-2-rhks。将此质粒导入到大肠杆菌BL21(DE3)中进行异源表达。在25 ℃和1 mmol/L异丙基-β-D-硫代吡喃半乳糖苷诱导条件下,组氨酸激酶融合蛋白(GST-RHK)获得成功表达,并具有催化活性。经谷胱甘肽琼脂糖亲和层析纯化,获得电泳纯的GST-RHK,其中纯化倍数为3.1,得率为19.5%。该蛋白大小约为72.75 kDa,Km、Vmax和Kcat值分别为20.92 μmol/L、0.17 μmol/(L·min)和1.4 min-1。野生型赤红球菌、组氨酸激酶基因增强株sdrhkE和组氨酸激酶基因敲减株sdrhkD在分别含有苯酚、甲苯、氯苯、异辛烷4 种有机溶剂的培养基中培养,菌株sdrhkD的生长情况都优于野生型赤红球菌,菌株sdrhkE的生长情况都低于野生型赤红球菌。本研究为进一步揭示赤红球菌SD3中组氨酸激酶涉及的信号转导途径与赤红球菌有机溶剂耐受性的关联机制提供一定参考依据。  相似文献

以三聚氰胺-甲醛树脂(MF)为囊材、聚磷酸铵(APP)和次磷酸铝(AHP)为芯材,制备出共微胶囊化阻燃剂M(A-A)。通过傅里叶变换红外光谱、扫描电子显微镜及溶解度测试等方法来表征MF的包覆效果;采用垂直燃烧测定仪、极限氧指数仪和锥形量热仪等设备考察M(A-A)对聚丙烯(PP)的阻燃效果;通过冲击和拉伸实验对复合材料的力学性能进行表征。结果表明,MF树脂成功包覆并有效提高了A-A的耐水性能;添加相同质量的M(A-A)和A-A,前者明显降低热释放速率(RHRR)和总热释放量(HTHR),对PP的阻燃效果更好。添加阻燃剂后,复合材料的冲击强度先提高后降低,经过微胶囊化处理的阻燃剂对材料的拉伸性能损伤更小。  相似文献

Chemotherapy is the mainstream treatment of anaplastic large cell lymphoma (ALCL). However, chemotherapy can cause severe adverse effects in patients because it is not ALCL‐specific. In this study, a multifunctional aptamer‐nanomedicine (Apt‐NMed) achieving targeted chemotherapy and gene therapy of ALCL is developed. Apt‐NMed is formulated by self‐assembly of synthetic oligonucleotides containing CD30‐specific aptamer and anaplastic lymphoma kinase (ALK)‐specific siRNA followed by self‐loading of the chemotherapeutic drug doxorubicin (DOX). Apt‐NMed exhibits a well‐defined nanostructure (diameter 59 mm) and stability in human serum. Under aptamer guidance, Apt‐NMed specifically binds and internalizes targeted ALCL cells. Intracellular delivery of Apt‐NMed triggers rapid DOX release for targeted ALCL chemotherapy and intracellular delivery of the ALK‐specific siRNA induced ALK oncogene silencing, resulting in combined therapeutic effects. Animal model studies reveal that upon systemic administration, Apt‐NMed specifically targets and selectively accumulates in ALCL tumor site, but does not react with off‐target tumors in the same xenograft mouse. Importantly, Apt‐NMed not only induces significantly higher inhibition in ALCL tumor growth, but also causes fewer or no side effects in treated mice compared to free DOX. Moreover, Apt‐NMed treatment markedly improves the survival rate of treated mice, opening a new avenue for precision treatment of ALCL.  相似文献

Ergothioneine has emerged as a crucial cytoprotectant in the pathogenic lifestyle of Mycobacterium tuberculosis. Production of this antioxidant from primary metabolites may be regulated by phosphorylation of Thr213 in the active site of the methyltransferase EgtD. The structure of mycobacterial EgtD suggests that this post-translational modification would require a large-scale change in conformation to make the active-site residue accessible to a protein kinase. In this report, we show that, under in vitro conditions, EgtD is not a substrate of protein kinase PknD.  相似文献

布鲁顿式酪氨酸激酶(Btk)和两面神激酶3(JAK3)均是自身免疫性疾病和血液系统恶性肿瘤的潜力靶标。以4-氯吡咯并嘧啶为原料,经7步反应合成了一系列7H-吡咯并[2,3-d]嘧啶-4-胺衍生物,其结构经1HNMR、13CNMR和MS(ESI)分析证实。体外依次考察了所得化合物分别对Btk和JAK3激酶的活性、以及对部分B淋巴细胞瘤细胞系的抗增殖活性。结果表明,四氢吡咯烷基取代的衍生物NB1~4对Btk(IC50<3 nmol/L)和JAK3(IC50<165 nmol/L)均能有效抑制,其余化合物只对Btk显示抑制效果;而且NB2和NB4对所测试的多种B细胞淋巴瘤细胞系的抗增殖活性均优于阳性对照泛激酶抑制剂依鲁替尼及Btk单靶点抑制剂。其中,对Jeko-1细胞的抑制能力最佳(IC50<3μmol/L),其次是OCI-LYI(IC50<4μmol/L)、SUDHL-6(IC50<6μmol/L)和HBL-1*(IC50<6.5μmol/L),最差为SUDHL-4细胞(IC50<11μmol/L)。这些发现可为开发B细胞淋巴瘤的多靶点药物提供新的思路。  相似文献

Vegetable soup (VS), a plant-based functional food, has been used as a traditional folk medicine and is attracting attention for its ability to enhance the immune response. β-Glucan, a well-established and effective immunomodulator, has synergistic effects when used in combination with some bioactive compounds. In the present study, we aimed to evaluate the synergistic immunomodulatory effects of the combination of VS and β-glucan on macrophage-mediated immune responses. β-Glucan was demonstrated to synergistically enhance the VS-stimulated immune response, including the production of interleukin-6, tumor necrosis factor-α, and nitric oxide, mainly through the mitogen-activated protein kinase pathway in macrophages. This paper describes research in areas where the occasional failure to meet the EC standard for lead in drinking water is due to the presence of particulate lead. A method is described to disinguish between soluble, particulate and colloidal lead in tap water, and characteristics are given of water sources which are most likely to suffer from this form of lead problem. At least two types of particulate (insoluble) lead in tap water have been identified, and some factors are discussed which cause the occurrence of both types. The vascularization of tissue-engineered bone is the key problem needed solving before application of tissue-engineered bone in clinical practice. Meanwhile, endothelial cells are the major and important source of seed cells in bone tissue engineering, and significant on promoting vascularization in tissue-engineered bone. Vascularization (namely angiogenesis) is a process mainly controlled by several angiogenic growth factors (VEGF, bFGF and MMP-2) which can be secreted by endothelial cells. Therefore, the research on the stimulations of SCPP to the secretion of the angiogenic growth factors from endothelial cells is very important. This study was performed to determine the ability of strontium-doped calcium polyphosphate (SCPP) to induce angiogenesis by detecting the protein secretion levels and mRNA expression of VEGF, bFGF and MMP-2 from cultured endothelial cells. As a control, we also researched the effect of HA on the mRNA expressions and protein secretion of angiogenic growth factors from cultured endothelial cells. We cultured endothelial cells with SCPP scaffolds containing various concentration of strontium and HA. The results obtained in the MTT and SEM tests indicated that endothelial cells on SCPP scaffold exhibited higher proliferation rate and were easy to get a good spread than them on CPP, the best state of growth and proliferation of cells could be observed on 8%SCPP. The results of ELISA demonstrated that the protein levels of VEGF, bFGF and MMP-2 from cultured endothelial cells increased with the increasing Sr doped in calcium polyphosphate in SCPP groups, the peaks appeared on 8%SCPP. All SCPP groups showed a better ability to stimulate the protein secretion of VEGF, bFGF and MMP-2 from endothelial cells relative to CPP group and HA group. The results of RT-PCR suggested that the 8%SCPP group exhibited a significantly higher mRNA expression of VEGF, bFGF and MMP-2 relative to CPP group and HA group. In conclusion, the results of this study demonstrated that 8%SCPP had obvious promotion for secretion and mRNA expression of angiogenic growth factors from cultured endothelial cells.  相似文献
